No,  there should be no comman between the statistics-type and the filter.
The comma is used to separate into separate columns in the output.

I think there is some issue with these stats just not working at all for
frame.xxx  fields, it could be something wrong with how we tap from "frame"
but have not had time to investigate.

For IP based traffic one can try using
-z "io,stat,100,AVG(ip.len)ip.len" instead

I think that should be

        -z "io,stat,100,AVG(frame.pkt_len),frame.pkt_len"

with a comma between the AVG() _expression_ and the filter.
