If you want a capture filter, try:
port 25 and not host 1.2.3.4 and not host 2.3.4.5
This will filter all traffic to tcp port 25, and exclude 1.2.3.4 and
2.3.4.5.

I would like to create a filter which would capture SMTP traffic, this is
easy enough but how would I exclude 2 IP addresses on our network from being
captured?
